You would likely have to put a LOT of revolutions to get the dots to line up again. It's not an every 2 turns thing. Pretty sure there are an odd number of teeth on the belt to cause it to 'walk' so the belt doesn't constantly contact the same place on the sprockets frequently.
Turning 720 is to make sure nothing is hitting inside the engine and for the tensioner to find it's normal extension. Check the plunger measurement, get it in spec and you should be OK.
